The Jackson Chamber of Commerce grew out of an earlier Commercial Club, an organization of merchants active in the city in the early 1900s. The Chamber of Commerce was chartered by Missouri in 1955, with a reported membership of 86 members. Membership has steadily increased to the current strength of 395.

The first chamber office was opened and staffed with a full-time secretary in 1974 at the intersection of highways 61 and 25. In October 1984, the Chamber of Commerce purchased the post office building at 125 E. Main St. (the current location). In 1998, the city of Jackson and the Chamber of Commerce entered into an agreement to assist in the funding of the Chamber for Economic Development and had to hire an executive director to manage the day-to-day operations. A valuable partnership was formed and continues to flourish between the city and the chamber.

The chamber's mission is to strengthen and promote member business, civic interest and enhance the community's quality of life. Jackson Chamber of Commerce partners include gold partners Bank of America and Regions Bank; silver partner Capaha Bank, Commerce Bank, First Midwest Bank, First Missouri State Bank, Montgomery Bank, The Bank of Missouri and the Cash-Book Journal newspaper.
