The Blue Devils (17-1) have never been 18-0, and paint-defending, layup-producing Georgetown wasn't about to let it happen Saturday, despite J.J. Redick matching his career high of 41 points for Duke.
The unranked Hoyas shot 61 percent, built a double-digit lead deep into the second half and nervously survived the frantic final minutes.
No. 2 Florida and (17-1) and No. 9 Pittsburgh (15-1) also suffered their first losses Saturday, leaving Division I without an unbeaten team.
Brandon Bowman scored 23 points for the Hoyas (12-4), who hadn't beaten a No. 1 team since an 85-69 win over St. John's on Feb. 27, 1985. The victory marks the first defining moment for the school under coach John Thompson III.
Shelden Williams scored a season-low four points for Duke.
Dane Bradshaw scored with 20 seconds left and Chris Lofton hit two free throws to seal it for Tennessee at home.
Lofton scored 29 points for Tennessee (12-3, 3-1).
Lamont Hamilton matched his career high with 24 points and St. John's pinned the first loss on visiting Pittsburgh. St. John's, which beat No. 17 Louisville 68-56 on Tuesday, scored the game's first 14 points and held a 31-19 halftime lead.
